Grass carp (Ctenopharyngodon idella), crucial to global inland aquaculture with a production of 5.8 million tones in 2020, faces significant challenges from hemorrhagic disease caused by grass carp reovirus (GCRV). Rapid mutations compromise current vaccines, underscoring the need for a deeper understanding of antiviral mechanisms to enhance molecular marker-assisted selection. This study investigates the role of Tripartite Motif (TRIM) family in the innate immune response of grass carp, focusing on TRIM103 from Ctenopharyngodon Idella (CiTRIM103), a member of the TRIM-B30.2 family, which includes proteins with the B30.2 domain at the N-terminus, known for antiviral properties in teleosts. CiTRIM103 bind to the outer coat proteins VP5 and VP7 of GCRV. This binding is theorized to strengthen the function of the RIG-I-like Receptor (RLR) signaling pathway, crucial for antiviral responses. Demonstrations using overexpression and RNA interference (RNAi) techniques have shown that CiTRIM103 effectively inhibits GCRV replication. Moreover, molecular docking and pulldown assays suggest potential binding interactions of CiTRIM103's B30.2 domain with GCRV outer coat proteins VP5 and VP7. These interactions impede viral replication, enhance RLR receptor expression, and activate key transcription factors to induce type I interferons (IFNs). These findings elucidate the antiviral mechanisms of CiTRIM103, provide a foundation for future Molecular genetic breeding in grass carp.

摘要:
The inconsistency between traditional morphological taxonomy and molecular phylogenetic data is a major issue that puzzles the study of fish classification and evolution. Although mitochondrial genes are commonly used in phylogenetic analyses to compare fish species, the mitochondrial evolution and diversity of Schistura are still not well understood. To better understand the evolution of Schistura, we sequenced the mitochondrial genome of Schistura fasciolata and compared it with other species of Schistura. A 16,588 bp circular mitochondrial genome of S. fasciolata was obtained and it contains 13 protein-coding, 22 transfer RNA, and two ribosomal RNA genes, and a non-coding control region. The gene arrangement in the mitochondrial genomes of all Schistura species was consistent. However, we also found that S. fasciolata was not monophyletic. Although mitochondrial genes can be effectively used for Schistura species identification, they may not be suitable for inferring the evolutionary process of Schistura species. These results provide support for the use of mitochondrial genes in identifying Schistura species, and also serve as a warning against mistakenly using them to evaluate the evolution process of Schistura species.

采用RT–PCR和cDNA末端快速扩增技术克隆草鱼(Ctenopharyngodon idellus)II型小肽转运载体(PepT2)基因全长cDNA序列,对其进行生物信息学和共线性分析,并采用实时荧光定量PCR技术检测...展开更多 采用RT–PCR和cDNA末端快速扩增技术克隆草鱼(Ctenopharyngodon idellus)II型小肽转运载体(PepT2)基因全长cDNA序列,对其进行生物信息学和共线性分析,并采用实时荧光定量PCR技术检测分析健康草鱼PepT2基因在不同组织中的表达情况。结果表明:草鱼PepT2基因的cDNA序列全长为2733 bp,包括开放阅读框2169 bp(编码722个氨基酸残基),5′非编码区82 bp和3′非编码区482 bp;草鱼PepT2基因结构含有20个外显子和19个内含子;预测蛋白相对分子质量为8.032×104,等电点为6.01,该蛋白具有与哺乳动物同源蛋白类似的12个螺旋跨膜结构,并且在跨膜区9和10之间有1个大的外环,跨膜区氨基酸高度保守,含有6个蛋白激酶C磷酸化位点和3个N–糖基化位点;预测的PepT2蛋白三级结构包含有18个α–螺旋和21个β–折叠;氨基酸序列同源性分析结果显示,草鱼PepT2氨基酸序列与斑马鱼(Danio rerio)的同源性最高,为86.65%,与其他所比较的物种的同源性则为52.83%~68.15%;系统进化树分析表明,草鱼与斑马鱼的亲缘关系最近;与斑马鱼相比,草鱼PepT2所在的染色体保留着大多数基因,具有保守的基因块;实时荧光定量表达分析表明,草鱼PepT2在肠道中的表达量最高,在肾脏中的表达量次之。收起

沉默信息调控因子2样蛋白3(SIRT3或Sirtuin3)是位于线粒体的主要去乙酰化酶,是机体脂代谢及抗氧化防御的重要调节者.SIRT3对脂肪酸β-氧化关键酶长链酰基CoA脱氢酶(LCAD)等进行脱乙酰化修饰调节脂代谢,还可活化AMPK通路调节脂代谢.SIRT3还可对抗氧化因子超氧化物歧化酶(Mn-SOD/SOD2)和异柠檬酸脱氢酶2(IDH2)的赖氨酸活化位点脱乙酰化增强抗氧化防御.论文总结SIRT3在脂代谢和抗氧化防御的调节作用,以期为揭示能量代谢障碍性疾病的发病机制提供启示,进而为探寻防治新靶标奠定理论基础.

摘要:
Fumonisins (Fums) are types of mycotoxin that widely contaminante feed material crops, and can trigger potential biological toxicities to humans and various animals. However, the toxicity of Fums on porcine blood vessels has not been fully explored. Fumonisin B1 (FB1) is the main component of Fums. Therefore, the aim of this study was to explore the effects of FB1 on the oxidative stress and tight junctions of the pig iliac endothelial cells (PIECs) in vitro. The results showed that FB1 reduced the viability of PIECs, increased the contents of lipid peroxidation product malondialdehyde (MDA), decreased the activities of antioxidant enzymes superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT) and thioredoxin reductase (TrxR), and decreased the level of glutathione (GSH). In addition, the barrier functions were destroyed, along with the down-regulations on Claudin 1, Occludin and ZO-1 and the increase of paracellular permeability. Thus, this research indicates that FB1 facilitates oxidative stress and breaks barrier functions to damage pig iliac endothelium cells. © 2019 by the authors. Licensee MDPI, Basel, Switzerland.

Fumonisins (Fums) are mycotoxins widely distributed in crops and feed, and ingestion of Fums-contaminated crops is harmful to animal health. The purpose of this study is to explore the effect of Fum B1 (FB1) on barrier functions of porcine intestinal epithelial cells, IPEC-J2, to clarify the intestinal toxicity of Fums in pigs. The results showed that the persistent treatment of FB1 significantly decreased the viability of IPEC-J2. Moreover, the expressions of Claudin 1, Occludin, Zonula Occluden-1 (ZO-1) on the messenger RNA (mRNA), and protein levels and MUC1 on the mRNA level were significantly inhibited after FB1 treatment, while the mRNA relative expression level of MUC2 was clearly increased. FB1 also enhanced the monolayer cell permeability of IPEC-J2. Importantly, FB1 promoted the expression of phosphorylated extracellular regulated protein kinase (p-ERK1/2). These data suggest that long-term treatment of FB1 can suppress IPEC-J2 proliferation, damage tight junctions of IPEC-J2, and regulate expression of mucins to induce the damage of barrier functions of porcine intestinal epithelial cells, which may be associated with the ERK1/2 phosphorylation pathway. © 2019 Wiley Periodicals, Inc.

摘要:
The barbel chub (Squaliobarbus curriculus) is a kind of small size commercial fish species that is widely spread in Asia and has shown significant resistance to disease. In this study, the full-length cDNA sequences of Toll-like receptor (TLR) 7 and 8 from S. curriculus, designated as ScTLR7 and ScTLR8, were cloned, and their differences in the structure and the responses to the grass carp (GCRV) infection and lipopolysaccharide stimulation were investigated. The full-length 3715 base pair (bp) cDNA of ScTLR7 contained a complete open reading frame of 3162 bp and encoded a putative polypeptide of 1053 amino acid residues. The full-length 4624 base pair (bp) cDNA of ScTLR8 contained a complete open reading frame of 3072 bp and encoded a putative polypeptide of 1023 amino acid residues. ScTLR7 and ScTLR8 consisted of N-terminal signal peptide, leucine-rich repeats (LRRs), and Toll/IL-1 Receptors domain. LRR motifs of ScTLR7 and ScTLR8 bend into horseshoe-like solenoid structure, while the number of LRRs between the two genes is different. Phylogenetic analysis showed that both the ScTLR7 and ScTLR8 were closely clustered with Ctenopharyngodon idellus and Megalobrama amblycephala. Quantitative real-time polymerase chain reaction analysis showed that the expression levels of ScTLR7 in S. curriculus were most abundant in the brain followed by the spleen and heart, and the lowest in the intestine. The highest expression level of ScTLR8 was observed in spleen and the lowest in the liver. After LPS stimulation, the relative expression levels of both ScTLR7 and ScTLR8 exhibited an overall trend of up-regulation. The expression levels of type I-IFN showed an overall trend of down-regulation at time points of 12, 24, 72 and 168 h compared to that of 6 h after LPS stimulation. Compared to 6 h post GCRV infection, the transcription level of ScTLR7 was up-regulated from 12 to 168 h, and transcription levels of ScTLR8, MyD88, and type I-IFN were firstly up-regulated from 12 to 72 h, and then down-regulated from 72 to 168 h. Correlation analysis showed that expression level of ScTLR7 in the spleen was significantly positively correlated with that of MyD88 (Pearson correlation coefficient: 0.909, P: 0.033), and a significantly positive correlation was also observed between expression levels of MyD88 and type I IFN (Pearson correlation coefficient: 0.962, P: 0.009), after GCRV stimulation. These results indicate that ScTLR7 and ScTLR8 may play important roles in the responses to the grass carp (GCRV) infection and lipopolysaccharide stimulation and trigger different downstream immune signal pathways.

Weanling stress and toxicosis, which are harmful to the health of pigs' intestines, are associated with oxidative stress. Quercetin (Que) is a polyphenolic compound that shows good anti-cancer, anti-inflammation and anti-oxidation effects. This study aimed to elaborate whether or not Que promotes IPEC-J2 (intestinal porcine enterocyte cells) proliferation and protects IPEC-J2 from oxidative damage. Thus, we examined the effects of Que on proliferation and H2O2-induced apoptosis in IPEC-J2. The results showed that Que increased IPEC-J2 viabililty, propelled cells from G1 phase into S phase and down-regulated gene levels of P27 and P21, respectively. Besides, H2O2-induced cell damage was alleviated by Que after different exposure times, and Que depressed apoptosis rate, reactive oxygen species (ROS) level and percentage of G1 phase cells and elevated the percentage of cells in G2 phase and S phase and mitochondrial membrane potential (Δψm) after IPEC-J2 exposure to H2O2. Meanwhile, Que reduced the value of Bax/Bcl-2 in H2O2 exposed cells. In low-degree oxidative damage cells, lipid peroxidation product malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were increased. In turn, Que could reverse the change of MDA content and SOD activity in low-degree damage cells. Nevertheless, catalase (CAT) activity was not changed in IPEC-J2 incubated with Que under low-degree damage conditions. Interestingly, relative expressive levels of the proteins claudin-1 and occludin were not altered under low-degree damage conditions, but Que could improve claudin-1 and occludin levels, slightly. This research indicates that Que can be greatly beneficial for intestinal porcine enterocyte cell proliferation and it protects intestinal porcine enterocyte cells from oxidation-induced apoptosis, and could be used as a potential feed additive for porcine intestinal health against pathogenic factor-induced oxidative damages and apoptosis. © 2018 by the authors.

摘要:
Purpose: To investigate the effect of Quercetin (Que) on inflammatory reaction in intestinal porcine enterocyte cells, IPEC-J2, induced by lipopolysaccharide (LPS). Methods: IPEC-J2 cells were pretreated with Que and then incubated with LPS. Cell viability, cell morphology, nitric oxide (NO) content, gene expression levels of interleukin-6 (IL-6) and interleukin-8 (IL-8), were then evaluated. Results: LPS incubation at 10 μg/mL for 24 h showed no effect on the viability and cell morphology of IPEC-J2, while Que pre-incubation significantly enhanced the viability of IPEC-J2 and improved cell morphology (p < 0.05). Que decreased the expression levels of pro-inflammatory cytokines IL-6 mRNA and IL-8 mRNA raised by LPS (p < 0.05). Interestingly, both LPS and Que demonstrated no influence on the release of NO. Conclusion: Pre-treatment of Que shows a positive effect on intestinal porcine enterocyte cells and inhibited porcine intestinal inflammation in vitro induced by LPS, but the mechanism may be not associated with NO-related signaling pathway. 为探究高锡对雏鸡肠道氧化应激的影响,试验将1日龄黄羽肉鸡120只随机分为4组,每组30只,对照组饲喂基础日粮,高锡1,2,3组日粮中分别添加400,800,1 200 mg/kg氯化亚锡,进行为期42 d的饲养试验,每天对雏鸡进行临床观察记录,并于雏鸡7,14,21,28,35,42日龄时称重,测定肠道丙二醛(MDA)含量和总超氧化物歧化酶(T-SOD)活性.结果表明:从21日龄起,高锡3组雏鸡体重显著或极显著低于对照组(P<0.05或P<0.01),且高锡3组雏鸡的空肠与回肠MDA含量显著或极显著高于对照组(P<0.05或P<0.01),高锡3组空肠T-SOD活性显著或极显著低于对照组(P<0.05或P<0.01).说明高剂量锡会阻碍雏鸡生长发育,并引起肠道氧化应激,从而破坏肠道抗氧化功能,对雏鸡的健康产生严重的危害.

动物生物化学是畜牧和水产相关专业的一门重要基础课程,如何让学生更好地掌握这门课程的内容,关系到学生后续专业课程的学习和理解,以及学校对相关专业人才的培养。文章在充分掌握动物生物化学课程的教学目标、特点和实际教学情况的基础上,以现代教育技术为手段,从改善教学条件、完善教学体系、加强实践培养、教学科研相结合等方面探讨对课程的改革,可以极大地提高了学生的兴趣,以及提出问题、分析问题、解决问题的能力和学习的主动性和积极性,同时可以全面提高课程的学习过程中学生对于课程的体验。

几种常见化合物对槲皮素稳定性的影响

摘要:
槲皮素是一种多酚羟基化合物,具有抗炎、抗氧化、提高动物生产性能、改善动物肉品质等功能。为探究生产或加工过程中常见化合物对槲皮素稳定性的影响,选取了不同浓度的氧化还原剂、金属化合物、糖类、柠檬酸等物质处理槲皮素6 h和24 h,并观察几种不同浓度的常见化合物对槲皮素浓度的影响。结果表明,过氧化氢和亚硝酸钠呈时间和浓度依赖性地降低槲皮素浓度(P〈0.05);钾离子、钙离子、钠离子、葡萄糖、蔗糖和柠檬酸不影响槲皮素浓度(P〉0.05);镁离子与槲皮素作用6 h,对其浓度无显著影响(P〉0.05),而作用24 h时,高浓度的镁离子显著降低了槲皮素溶液浓度(P〈0.05);铝离子和铜离子作用6 h和24 h均能显著地降低槲皮素浓度(P〈0.05)。表明氧化剂、还原剂、镁离子、铜离子和铝离子会影响槲皮素稳定性,而钾离子、钙离子、钠离子、葡萄糖、蔗糖和柠檬酸不会影响槲皮素稳定性。

摘要:
目的:评价研究性学习教学方法在中兽医学实验教学中的应用效果。方法:将中兽医学的实验教学内容设计为研究性学习教学模式,以2012级动物医学本科专业54名学生为研究对象,随机分为对照组与实验组,对照组采用传统教学法,实验组采用研究性学习教学法,从教学原则、设计思路、教学内容、教学方法、教学手段等方面进行探讨,以实验考试与问卷调查的方式进行教学效果评价。结果:在提高学生自学能力、创新能力等各项评价指标上,研究性学习教学法与传统教学法相比,在统计学上具有显著性的差异(P﹤0.05);实验组的实验考试平均成绩明显高于对照组(P﹤0.05)。结论:研究性学习教学方法能较好地培养学生的综合素质,亦为其他实验课程教学提供借鉴。

摘要:
A full-length cDNA clone encoding grass carp (Ctenopharyngodon idellus) α1-microglobulin/bikunin precursor (Ci-AMBP) was isolated by subtracted differential hybridization screening from a liver cDNA library. The deduced amino acid sequence shared approximately 50% sequence identity with its mammalian counterparts, but more than 90% identity with another fish species. AMBPs are the precursors of the plasma glycoproteins α1-microglobulin (α1m) and bikunin. Both peptide structures and their chromosomal organization were well conserved in Ci- AMBP. The α1m and bikunin polypeptides are separated by the typical tetrapeptide R-A-R-R that provides an endoproteolytic cleavage site for maturation. The genetic organization of domains and functional motifs indicated that Ci-AMBP is a typical member of the lipocalin and Kunitz-type protease inhibitor superfamilies. Expression of the Ci-AMBP gene in different tissues/organs was evaluated using semi-quantitative RT-PCR and, in contrast to the restricted expression in other species, transcripts were detected in a wide range of tissues. The most abundant expression occurred in the secretory organs, which supports the roles of α1m and bikunin in the immune response to diseases and in the stress response. © 2017, Genetics Society of Japan. 为探究赤眼鳟(Squaliobarbus curriculus) Toll样受体3(Toll-like receptor 3, ScTLR3)基因是否参与抗病毒免疫反应,实验运用RACE技术,克隆得到ScTLR3基因cDNA全长序列,并进行了生物信息学分析;通过Real- Time qPCR技术,检测了ScTLR3 mRNA在健康赤眼鳟10个组织中的分布以及感染草鱼呼肠孤病毒(GCRV)后肝脏、脾脏、体肾和头肾中的表达特征。结果表明: ScTLR3基因cDNA序列全长4043 bp,包括5'-非编码区(UTR) 216 bp,开放阅读框(ORF)2715 bp和3'-UTR 1112 bp; ScTLR3共编码904个氨基酸残基,推导的蛋白分子量102.67 kD,理论等电点8.76; SMART结构域预测显示, ScTLR3由N端的信号肽(SP)、富亮氨酸结构域(LRRs)、跨膜结构域(TM)和C端的Toll/白介素-1受体结构域(TIR)组成。实时荧光定量结果显示, ScTLR3 mRNA在检测的各组织中均有表达,肝脏中的相对表达量极显著高于其他组织(P<0.01);感染GCRV后,肝脏、脾脏、体肾和头肾组织中ScTLR3 mRNA均上调表达,肝脏、脾脏和体肾组织中的相对表达量在24 h达到峰值,分别为对照组的5倍、7倍和6倍。研究表明, ScTLR3具有TLRs家族基因的典型结构特征,并能被GCRV诱导表达,推测其在赤眼鳟抗GCRV入侵免疫反应中发挥了重要作用。

鸡屎藤是我国一种传统的药食同源性植物,含环烯醚萜苷类、挥发油类、黄酮类等化学成分,有镇痛、抗炎、抗氧化、保肾和抗菌等药理作用。近年,鸡屎藤的化学成分、药理活性机制以及活性成分的提取工艺等研究较为深入。笔者通过查阅近年文献,对鸡屎藤的化学成分、提取工艺与药理作用进行了综述,以期为鸡屎藤的深入研究与资源利用开发提供参考。

摘要:
为构建一种简便、高效的槲皮素原料药含量的测定方法,采用紫外分光光度法对槲皮素原料药进行含量测定,并进行方法学考察.结果表明,紫外分光光度法在374 nm处测定的槲皮素原料药平均含量为97.97%,标准曲线方程为y=0.007 4x-0.006 7,R2 =0.999 7,在20.8 μg~140 μg范围内有良好的线性关系,平均加样回收率为100.88%,RSD为0.96%(n=6),其精密度较好,符合方法学要求.说明该法适用于槲皮素原料药含量测定,是一种经济、简便与高效的质量分析方法.

槲皮素(quercetin,QCT)是一种主要存在于水果、蔬菜、草本植物中的黄酮类化合物,近年来广受关注。本研究就2007—2016年QCT的文献分布情况进行了初步分析,发现发文量总体呈递增趋势;并对其药代动力学以及抗癌、抗肥胖、抗阿尔茨海默病、抗抑郁和免疫调节等生物活性的研究进展进行了综述,以期为QCT的深入研究和进一步开发利用提供参考。

摘要:
The aim of the study was to investigate the grass carp hemorrhagic infection pathway and its key-related genes. Grass carp reovirus (GCRV) might cause hemorrhagic disease in grass carps. Healthy grass carp fingerlings (N = 60) were divided into control and infected groups. Fish in the control group were intraperitoneally (ip) injected with 0.6% fish physiological saline, the infected group received 5,000,000 50% tissue culture infective doses of GCRV 873 standard strain, a double-stranded RNA (dsRNA) virus strain, ip, in 0.5 mL. Illumina HiSeq™ 2000 was used for transcriptome sequencing, and real-time polymerase chain reaction (PCR) used to detect complement factors II (C2), III (C3), and V (C5), profibrinolysin (PLG), and coagulation factor II (F2) expression. A total of 2,722,223 reads were detected in the control group, and 2,751,111 in the infected group. Among 11,023 unigenes obtained after transcriptome assembly, 10,021 unigenes were significantly differentially expressed. Gene ontology and KEGG analysis, a collection of databases dealing with genomes and biological pathways, were performed to classify unigenes into functional categories, to understand gene function and identify regulatory pathways. Real-time PCR analysis showed that C2, C3, C5, PLG, and F2 expression levels were down-regulated, confirming results of pathway-enrichment analysis. This is the first application of highthroughput sequencing technology to investigate the in vivo effects of GCRV, on genes and pathways involved in the immune response to infection in grass carp. © FUNPEC-RP.
